AS A RESULT OF OUR PREVIOUS PROJECT, THREE TWO COMPONENT (TC) SYSTEMS OF LACTOBACILLUS CASEI HAVE BEEN SHOWN TO BE INVOLVED IN THE RESPONSE OF THIS ORGANISM AGAINST DIFFERENT STRESS CONDITIONS AND GENES WITH ALTERED EXPRESSION IN TC-DEFICIENT MUTANTS ARE BEING IDENTIFIED, THIS PROJECT INTENDS TO EXTEND OUR KNOWLEDGE OF THE PHYSIOLOGICAL ROLE OF THESE TC SYSTEMS BY DETERMINING THE STRUCTURE OF THEIR REGULATORY NETWORKS, TO THIS END, DNA BINDING SITES RECOGNIZED BY THE CORRESPONDING RESPONSE REGULATORS WILL BE DETERMINED BY A COMBINATION OF BIOINFORMATIC AND MOLECULAR BIOLOGY METHODS IN ORDER TO DETERMINE GENES UNDER DIRECT CONTROL OF RRS, SUBSEQUENTLY, CRITICAL NODES IN THE REGULATORY NETWORKS WILL BE IDENTIFIED BY GENE INACTIVATION AND COMPARATIVE GLOBAL GENE EXPRESSION ANALYSES, IN ADDITION, SYSTEM TC17, INVOLVED IN THE REGULATION OF L-MALIC ACID METABOLISM HAS BEEN CHOSEN FOR ITS UNIQUE PROPERTIES FOR A DETAILED CHARACTERIZATION AT THE MOLECULAR LEVEL OF THE INTERACTIONS BETWEEN THE HISTIDINE-KINASE AND RESPONSE REGULATOR, FINALLY, THE ROLE OF TC SYSTEMS IN THE RESPONSE TO PHENOLIC COMPOUNDS WILL BE ALSO ADDRESSED IN THIS PROJECT, TO THIS END, TC SYSTEMS INVOLVED IN RESISTANCE TO SELECTED PHENOLIC COMPOUNDS WILL BE IDENTIFIED BY GROWTH ASSAYS WITH OUR COLLECTION OF MUTANTS DEFICIENT IN EACH TC SYSTEM, SUBSEQUENTLY, GLOBAL GENE EXPRESSION ASSAYS WILL BE CARRIED OUT IN ORDER TO IDENTIFY GENES INVOLVED IN THE RESPONSE AND PUTATIVELY UNDER CONTROL OF THE TC SYSTEMS IDENTIFIED,
