OUR UNDERSTANDING OF THE REGULATION OF GENOMIC PROCESSES HAS INCREASED TREMENDOUSLY OWN TO THE IDENTIFICATION AND FUNCTIONAL CHARACTERIZATION OF FACTORS AND MECHANISMS INVOLVED IN THE EPIGENETIC REGULATION OF CHROMATIN STRUCTURE AND FUNCTION, IN THIS REGARD, OUR WORK AIMS TO THE IDENTIFICATION OF NOVEL EPIGENETIC FACTORS AND MECHANISMS, IN RECENT YEARS, WE HAVE FOCUSED OUR STUDIES TO THE ANALYSIS OF LINKER HISTONES H1 AND HP1 PROTEINS AS EPIGENETIC REGULATORS, AS WELL AS TO THE IDENTIFICATION AND FUNCTIONAL CHARACTERIZATION OF NEW CENTROMERIC COMPONENTS, IN THE LAST YEARS, OUR MAIN ACHIEVEMENTS HAVE BEEN: 1) LINKER HISTONES H1 FUNCTIONS, WE HAVE UNVEILED THE ESSENTIAL ROLE OF SOMATIC DH1 IN THE MAINTENANCE OF GENOME STABILITY (BAYONA, CASAS-LAMESA ET AL,, (2017) NATURE COMMUN, 8: 283; VUJATOVIC ET AL,, (2012) NUCLEIC ACIDS RES, 40: 5402), WE HAVE DETERMINED THE PATTERN OF POST-TRANSLATIONAL MODIFICATIONS (PTMS) OF DH1 (BONET ET AL,, (2012) J PROTEOMICS, 75: 4124), WE HAVE ALSO IDENTIFIED A NOVEL GERMLINE SPECIFIC LINKER HISTONE, DBIGH1, AND UNCOVERED ITS CRUCIAL FUNCTIONS IN GERM STEM CELL (GSC) LINEAGE DIFFERENTIATION AND EARLY EMBRYO DEVELOPMENT (CARBONELL ET AL,, (2017) CELL REP, 21: 3178; PEREZ-MONTERO ET AL,, (2013) DEV CELL, 26: 578); 2) HP1 PROTEINS, WE HAVE IDENTIFIED A NOVEL COMPLEX FORMED BY THE HP1B/C PROTEINS, THE TRANSCRIPTION FACTORS WOC AND ROW, AND THE UBIQUITIN RECEPTOR DDSK2, WHICH REGULATES RNAPOL II PAUSING AT PROMOTERS OF TARGET GENES (KESSLER ET AL,, (2015) NATURE COMMUN, 6:7049; FONT-BURGADA ET AL,, (2008) GENES DEV, 22: 3007); 3) CENTROMERE ASSEMBLY AND FUNCTION, WE HAVE DETERMINED THE ESSENTIAL ROLE OF PROTEOLYSIS IN THE ASSEMBLY OF CENTROMERIC CHROMATIN (MORENO-MORENO ET AL,, (2018) NUCLEIC ACIDS RES, 2ND ROUND OF REVIEW; MORENO-MORENO ET AL,, (2011) CURR BIOL, 21: 1488; MORENO-MORENO ET AL,, (2006) NUCLEIC ACIDS RES, 34: 6247), WE HAVE ALSO IDENTIFIED SEVERAL NOVEL CENTROMERE ASSOCIATED PROTEINS (MEDINA-GIRO, TORRAS-LLORT ET AL, UNPUBLISHED), IN THIS PROPOSAL, WE AIM TO COMPLETE AND EXTEND THESE STUDIES TO COVER NEW ASPECTS RELATED TO GENERAL QUESTIONS OF BROAD INTEREST IN THE FIELD THAT REMAIN POORLY UNDERSTOOD, SUCH AS THE TRIDIMENSIONAL ORGANIZATION (3D) OF CHROMATIN AND, SECOND, THE COORDINATION OF NUCLEAR EVENTS DURING MITOSIS, BELOW IS A BRIEF DESCRIPTION OF THE SPECIFIC OBJECTIVES TO BE ADDRESSED IN THIS PROPOSAL:A) LINKER HISTONES H1 FUNCTIONSA1) ANALYSIS OF THE CONTRIBUTION OF DH1 TO R-LOOPS DYNAMICSA2) FUNCTIONAL ANALYSIS OF DH1 PTMSA3) ANALYSIS OF THE INTERPLAY BETWEEN THE SOMATIC DH1 AND THE GERMLINE SPECIFIC DBIGH1 VARIANTSB) 3D CHROMATIN ORGANIZATIONB1) ANALYSIS OF THE CONTRIBUTION OF LINKER HISTONES H1 TO 3D CHROMATIN ORGANIZATIONB2) ANALYSIS OF THE 3D CHROMATIN ORGANIZATION OF HP1B/C-COMPLEX TARGET GENESC) CENTROMERE ASSEMBLY AND FUNCTION: THE COORDINATION OF MITOTIC EVENTSANALYSIS OF THE CONTRIBUTION OF BAF TO THE COORDINATION OF CHROMOSOME SEGREGATION AND POST-MITOTIC NE ASSEMBLY, EPIGENÉTICA\CROMATINA\ORGANIZACIÓN 3D DE LA CROMATINA\CENTRÓMERO\ESTABILIDAD GENÓMICA\HISTONA H1\R-LOOPS\HP1\BAF
