CELL COMPARTMENTATION IS ESSENTIAL FOR ORGANISMAL FORM AND FUNCTION. WHILE OUR UNDERSTANDING OF THIS BIOLOGICAL FEATURE HAS SIGNIFICANTLY ADVANCED, NUMEROUS QUESTIONS REMAIN UNADDRESSED. OUR GOAL IS TO COMBINE OUR COMPLEMENTARY EXPERTISE TO REVEAL THE MECHANISMS BY WHICH CARGOES (BOTH PROTEINS AND LIPIDS) ARE TARGETED TO SPECIFIC COMPARTMENTS AND CELLS EXTERIOR, WHICH IN TURN DETERMINES THEIR FATE AND FUNCTION. AN ILLUSTRATIVE EXAMPLE IS LIPID PARTICLES FORMING WITHIN THE BILAYER OF ENDOPLASMIC RETICULUM (ER) MEMBRANE: THEIR RELEASE INTO THE CYTOPLASM NUCLEATES CELLULAR LIPID DROPLETS (LDS- SOURCE OF ENERGY), WHILE THEIR RELEASE INTO THE ER LUMEN FORMS CHYLOMICRONS (SECRETED TO HELP CLEAR CIRCULATING CHOLESTEROL AND DIETARY LIPIDS). WHAT CONTROLS LD AND CHYLOMICRON SYNTHESIS, DYNAMICS AND EXPORT (MALHOTRA AND POL)? NUMEROUS ANALOGOUS TRAFFICKING QUESTIONS REMAIN UNADDRESSED. WHAT IS THE MORPHOLOGICAL ORGANIZATION OF MEGACARRIERS NEEDED TO EXPORT BULKY CARGOES SUCH AS CHYLOMICRONS AND COLLAGENS FROM ER (MALHOTRA AND MENARGUEZ)? HOW IS THE SPECIFIC SECRETORY PATHWAY OF EXOSOMES REGULATED AT EARLY STAGES IN MULTIVESICULAR BODIES FOR LONG-DISTANCE INTER TISSUE COMMUNICATION (DEL POZO)? HOW ARE SECRETORY CARGOES SORTED IN THE GOLGI TO ENDOSOMAL ROUTES, AND PACKED INTO SPECIFIC CARRIERS FOR EXPORT TO SPECIFIC MEMBRANE DOMAINS (ALONSO)? HOW IS THE GOLGI COMPLEX ORGANIZED AND HOW DOES IT TARGET CARRIERS ALONG THE CYTOSKELETON (MALHOTRA, MENARGUEZ AND RIOS)? HOW ARE CARGOES TARGETED FOR INTERNALIZATION SORTED AND PACKED INTO SPECIFIC CARRIERS (GELI, ENRICH)? HOW ARE THESE EVENTS REGULATED AND INTEGRATED WITH OTHER CELL FUNCTIONS SUCH AS ECM REMODELING BY SPECIFIC STRUCTURES SUCH AS THE CYTOSKELETON AND CAVEOLAE (POZO, BELMONTE, RIOS, ENRICH, GELI). IN CONCLUSION, THE RESEARCHERS APPLYING TO THIS PROJECT HAVE COMMON SCIENTIFIC INTERESTS AND OBJECTIVES THAT COULD BE TACKLED AND ACHIEVED IN A MORE COMPETITIVE WAY IF WE SHARE KNOWLEDGE, TECHNOLOGIES, REAGENTS AND PERSONNEL.
