Descripción del proyecto
HIGH THROUGHPUT METHODS CURRENTLY AVAILABLE FOR SEARCHING GENES RELATED WITH ANY TRAITS HAVE INTRINSIC FLAWS BECAUSE OF THEIR DATA DRIVEN NATURE, THESE FLAWS RESULT IN THE PRESENCE OF A LARGE AMOUNT OF FALSE POSITIVES, PARTIALLY DUE TO THE ABSENCE OF A-PRIORY HYPOTHESIS, THIS PROPOSAL AIMS AT OVERCOMING THIS PROBLEM BY INJECTING NEW INFORMATION -SEMANTIC SIMILARITY AND PROTEIN INTERACTION- ON DATA DRIVEN HIGH THROUGHPUT METHODS, THIS TEXT PROPOSES NOVEL TOOLS TO MODEL THE NETWORK OF PROTEINS RELATED TO SPECIFIC PATHWAYS AND COMPUTING THEIR POSSIBLE RELATIONSHIP WITH THE DISEASE EMPLOYING SEMANTIC COMPUTING, GRAPH ANALYSIS AND INFORMATION THEORY, RESEARCH WILL COVER SPECTRAL CLUSTERING ON LARGE DISTANCES MATRICES, THE USE OF ALPHA-DIVERGENCES TO MEASURE GENETIC AND SEMANTIC SIMILARITY AND GRAPH THEORY TECHNIQUES, ALL CODE WILL BE ADAPTED TO DISTRIBUTED COMPUTING ON A CLUSTER AND RELEASED UNDER OPEN SOURCE LICENSE IF NOT SUITABLE FOR AN INTELLECTUAL PROTECTION PROCESS, THE METHODS WILL BE APPLIED TO SUDDEN DEATH PHENOMENA (DUE TO CARDIOVASCULAR DISEASE) AND DEPTH OF ANESTHESIA SENSITIVITY TO OPIATES, THE CONTRIBUTION OF THE ¿OMICS SUBPROJECT WILL BE IDENTIFYING COMPUTATIONALLY THE MOLECULAR PATHWAYS THAT ARE RELATED WITH MYOCYTES CONTRACTION AND REGULATION OF CARDIAC RHYTHM (MAINLY IN LONGQT AND BRUGADA SYNDROME) AND THE LOCAL INTERACTION PROTEIN NETWORK OF THE MU OPIOID PEPTIDE (MOP) AND GABAA RECEPTORSAS THE NUMBER OF ANNOTATIONS ON GENETIC DATABASES HAS INCREASED DRAMATICALLY ON THE PAST YEARS, THERE IS THE REASONABLE ASSUMPTION THAT FOR BOTH APPLICATIONS (CARDIOVASCULAR DISORDERS AND THE EFFECT OF CERTAIN POLYMORPHISMS ON THE DEPTH OF ANESTHESIA) THERE WILL BE SUFFICIENT ANNOTATION DATA IN GENE ONTOLOGY AND PROTEIN-PROTEIN INTERACTION DATABASES IN ORDER TO PERFORM A COMPUTATIONAL STUDY OF THE RELATED PROTEIN NETWORKS, THE TECHNOLOGY THAT BETTER EXPLOITS THIS NEW ANNOTATION WILL BE BEST POSITIONED IN NEXT FUTURE FOR FURTHER COMMERCIAL AND MEDICAL DEVELOPMENTS,THE MAIN PROPOSED OBJECTIVES IN THIS SUBPROJECT WILL BE TO: 1) BUILD PROTEIN-PROTEIN INTERACTIONS NETWORK FORM ONLINE DATABASES2) ENRICH THE PROTEIN NETWORK STRUCTURE WITH INFORMATION FROM GENE ONTOLOGIES 3) DEFINE DISTANCES BETWEEN PROTEINS GIVEN THE INTERACTION NETWORK, AND BETWEEN PROTEINS AND SEMANTIC TERMS THROUGH THE INTERACTION NETWORK AND ONTOLOGY INFORMATION EMPLOYING INFORMATION THEORY4) ASSIGN PROPERTIES TO THE NETWORK NODES RELATED TO GENETIC VARIABILITY, THROUGH CONSERVATION PROPERTIES AND GENE REGULATION INFORMATION5) PREDICT GO LABELS FROM PROTEIN PRIMARY SEQUENCES AND BENCHMARK THE PREDICTION THROUGH UNIPROT DATABASE6) BUILD AN WEB INTERFACE EXPOSING THE TOOLS DEVELOPED IN THE PROJECT7) APPLY THE FRAMEWORK TO THE LOCAL INTERACTOME RELATED WITH LONGQT SYNDROME, 8) APPLY THE FRAMEWORK TO DESCRIBE THE LOCAL INTERACTOME RELATED WITH THE INDIVIDUAL SENSITIVITY TO OPIATES IN ANALGESIA PROBLEMS (STARTING FROM OPRM1 GENE), 9) TO APPLY AND VALIDATE THE PROPOSED METHODS WITH PLATELET PATHWAYS PERFORMED BY ELECTROPHORESIS AND MALDI-TOF ANALYSIS,10) PROTECT THE OUTCOME OF THE PROJECT THROUGH INTELLECTUAL PROPERTY PROCESS OR PROPER LICENSING ANALISIS DE REDES DE INTERACCIONES PROTE\PRIORIZACION DE GENES\ONTOLOGIAS\DESORDENES CARDIOVASCULARES\BRUGADA SYNDROME\LONG-QT SYNDROME\MONITORIZACION DE ANESTESIA\APROXIMACION OMICA
