The regulation of gene expression at the level of transcription is critical during development and growth of an organism. Specifically, regulation of embryonic development depends on complex gene networks. At the transcription level, embryonic segmentation relies on transcription factor gradients, sequence-specific DNA-binding proteins and regulatory DNA sequences such as enhancers. These enhancers recruit transcriptional activators, resulting in the recruitment of RNA polymerase II and the general transcription factors to the core promoter. The core promoter is the ultimate target of a multitude of transcription factors that control the initiation of transcription.
In search of the biological significance of specific core promoter elements, I discovered that the core promoters of many developmentally important genes, particularly the Hox genes (which are key regulators of the development of the embryonic body plan), contain functional DPE motifs (Juven-Gershon, T., Hsu, J.-Y. and Kadonaga, J.T. (2008) Caudal, a key developmental regulator, is a DPE-specific transcriptional factor Genes and Development, 22, 2823-2830).
My goal is to decipher the complex regulation of eukaryotic gene expression. My focus is on the contribution of the core promoter to transcriptional regulation of gene networks. My lab studies transcription of DPE-containing genes from different aspects: examination of how widespread is the use of DPE in diverse developmental pathways and gene regulatory networks; discovery of the basal transcription factors that are needed to enable DPE transcription; and elucidation of the critical mechanisms of core-promoter-specific transcription.
The study of transcription and core promoter regulation is important for the understanding of development and differentiation. New insights into the regulation of gene expression under normal growth conditions will improve our understanding of abnormal regulation of gene expression, which leads to human diseases.
