THE DESIGN OF NEW STRATEGIES FOR THE SUSTAINABLE MANAGEMENT OF PLANT BACTERIAL DISEASES, AS WELL AS FOR THE ESTABLISHMENT OF SUCCESSFUL PLANT BREEDING PROGRAMS, REQUIRES IDENTIFICATION AND MOLECULAR CHARACTERIZATION OF BACTERIAL GENES INVOLVED IN THE INTERACTION WITH SPECIFIC PLANT HOSTS, P, SAVASTANOI PV, PHASEOLICOLA (PPH) AND PV, SAVASTANOI (PSV) ARE TWO PHYLOGENETICALLY RELATED MODEL ORGANISMS THAT PRODUCE BLIGHTS IN HERBACEOUS HOSTS OR TUMORS ON WOODY PLANTS, RESPECTIVELY, THE STUDY OF VIRULENCE IN BACTERIA OF THE P, SYRINGAE COMPLEX, INCLUDING P, SAVASTANOI PATHOVARS, HAS BEEN REVOLUTIONIZED BY GENOMICS, WITH THE SEQUENCING OF THE GENOMES OF THREE STRAINS, INCLUDING PPH 1448A, IN THIS PROJECT, WE UNDERTAKE THE IDENTIFICATION OF P, SAVASTANOI VIRULENCE GENES BY ANALYZING THE COMPLETE GENOME OF PPH 1448A AND A DRAFT GENOME OF PSV NCPPB3335 THAT IS BEING SEQUENCED BY A CONSORTIUM OF FOUR SPANISH GROUPS, AS WELL, WE PROPOSE TO FOLLOW A FUNCTIONAL COMPARATIVE GENOMIC APPROACH IN ORDER TO GAIN INSIGHTS INTO HOST-SPECIFIC VIRULENCE AND PATHOGENICITY DETERMINANTS FOR HERBACEOUS AND WOODY HOSTS, AS WELL AS THEIR EVOLUTION IN THE BACTERIAL POPULATION, THE PROJECT IS ORGANIZED AS TWO COORDINATED SUBPROJECTS LEAD BY TWO GROUPS THAT ARE SPECIALISTS IN EITHER THE PPH-BEAN (UNIVERSIDAD PUBLICA DE NAVARRA) OR THE PSV-OLIVE (UNIVERSIDAD DE MALAGA) PATHOSYSTEM, WHICH WILL STREAMLINE THE COMPARATIVE ANALYSES, THE OBJECTIVES OF THIS PROJECT ARE: 1) ANALYSIS AND ANNOTATION OF THE DRAFT GENOME OF P, SAVASTANOI PV, SAVASTANOI NCPPB3335, 2) COMPLETION AND CLOSURE OF THE SEQUENCE OF THE THREE PLASMIDS COMPLEMENT OF NCPPB3335 AND EVALUATION OF THEIR ROLE IN VIRULENCE, AND 3) COMPARATIVE HOST RANGE ANALYSIS AND CHARACTERIZATION OF PATHOGENICITY AND VIRULENCE GENES AMONG PPH AND PSV STRAINS, THROUGH THE STRUCTURAL AND FUNCTIONAL COMPARISON OF DIVERSE VIRULENCE GENES AND THEIR ROLE IN HOST RANGE DEFINITION, THE GROUPS OF GENES THAT WILL BE ANALYZED INCLUDE GENES BELONGING TO THE ESSENTIAL PLASMID-BORNE PATHOGENICITY ISLAND OF PPH, GENES IDENTIFIED BY GENOMIC ANALYSIS, AND PSV GENES REQUIRED FOR FULL GROWTH IN OLIVE PLANTS PREVIOUSLY IDENTIFIED BY SIGNATURE TAGGED MUTAGENESIS, Bacterias fitopatogenas\mecanismos de virulencia\Pseudomonas syringae\plasmidos
