REPRODUCTIVE PERFORMANCE IS ONE OF THE MOST IMPORTANT FACTORS AFFECTING DAIRY FARM PROFITABILITY AND THE NATIONAL ECONOMY, AS WELL AS THE LIVING STANDARDS OF RURAL AND URBAN SOCIETIES, COMPROMISED FERTILITY IN MODERN-DAY CATTLE-PRODUCTION SYSTEMS IS OFTEN CONSIDERED A FAILURE OF REPRODUCTIVE PHYSIOLOGY THAT MUST BE FIXED, REPRODUCTIVE SUCCESS DEPENDS ON A CHAIN OF WELL-SYNCHRONIZED EVENTS THAT INVOLVE MANY INTERACTING PHYSIOLOGICAL ENTITIES (E,G,, HYPOTHALAMUS, PITUITARY, OVARY, OOCYTE, FOLLICLE, UTERUS, AND FETUS), SUCH COMPLEXITY MAKES IT DIFFICULT TO PINPOINT THE KEY PHYSIOLOGICAL COMPONENTS GOVERNING REPRODUCTIVE SUCCESS, BUT ALSO TO QUANTIFY WHAT IMPACT THEIR IMPROVEMENT INDIVIDUALLY WOULD HAVE ON OVERALL FERTILITY, THUS, STUDIES ON PHYSIOLOGICAL MECHANISMS AND INTERACTIONS IN THE MATERNAL REPRODUCTIVE TRACT (OVIDUCT AND UTERUS) ARE ESSENTIAL, SUCH STUDIES WILL HELP ADVANCE OUR KNOWLEDGE OF MECHANISMS RELATED TO EARLY EMBRYO DEVELOPMENT AND WILL SUPPORT THE IMPROVEMENT OF ASSISTED REPRODUCTIVE TECHNOLOGIES INCLUDING IN VITRO EMBRYO PRODUCTION THAT SEEK TO MIMIC PHYSIOLOGICAL CONDITIONS AND GENERATE GOOD QUALITY EMBRYOS, MOREOVER, THESE ADVANCES COULD HELP IMPROVE FERTILITY TREATMENTS IN HUMANS AND DOMESTIC ANIMALS TO INCREASE THE EFFICIENCY OF PRODUCTION AND BREEDING SCHEMES, BASED ON THAT THE GLOBAL OBJECTIVE OF THE CURRENT PROPOSAL IS THE MOLECULAR CHARACTERIZATION OF OVIDUCTAL, UTERINE AND EMBRYO/CONCEPTUS EVS AS MODULATORS OF EMBRYO-MATERNAL CROSSTALK IN CATTLE AND THEIR POTENTIAL IMPLICATIONS FOR THE PREGNANCY SUCCESS, TO REACH THIS OBJECTIVE, THE FOLLOWING SPECIFIC OBJECTIVES WILL BE ADDRESSED: OBJECTIVE 1; ANALYZE THE SECRETION/COMPOSITION OF THE OVIDUCTAL AND UTERINE EVS IN RESPONSE TO THE HORMONAL PROFILE OF THE COW AT 4 DIFFERENT STAGES OF ESTROUS CYCLE (METESTRUS: DAY 1-4, PRESENCE OF AN EARLY CL; EARLY DIESTRUS: DAY 4-8, PRESENCE OF FUNCTIONAL CL; LATE DIESTRUS: DAY 8-17, PRESENCE OF MATURE CL; PROESTRUS: DAY 17-21, REGRESSED CL, FOLLICULAR PHASE); OBJECTIVE 2, SCRUTINIZE THE MOLECULAR AND PROTEIN CONTENT OF THE OVIDUCTAL AND UTERINE EVS IN VIVO IN RESPONSE TO THE EMBRYO PRESENT AT KEY STAGES OF DEVELOPMENT (DAY 3: 8-16 CELL; DAY 7: BLASTOCYST; AND DAY 15: ELONGATED CONCEPTUS); OBJECTIVE 3, SCRUTINIZE THE MOLECULAR AND PROTEIN CONTENT OF EVS PRODUCED IN VITRO USING OVIDUCTAL AND UTERINE EXPLANTS IN RESPONSE TO AN EARLY EMBRYO/CONCEPTUS (DAY 3: 8-16 CELL; DAY 7: BLASTOCYSTS; AND DAY 15: ELONGATED CONCEPTUS); OBJECTIVE 4, EVALUATE THE POTENTIAL CAPACITY OF IN VIVO OR IN VITRO PRODUCED EVS (OBJECTIVE 2 AND 3) TO EMBRYO DEVELOPMENT IN VITRO LEADING TO SUCCESSFUL PREGNANCY OUTCOME AFTER TRANSFER TO RECIPIENTS, CHARACTERIZATION OF EVS FROM BOVINE OF AND UF AND THEIR RELATIONSHIP WITH ESTRUS CYCLE AND KEY STAGES OF EMBRYO DEVELOPMENT IN VIVO AND IN VITRO WILL GIVE US RELEVANT INFORMATION ON THE MECHANISMS OF MATERNAL-EMBRYONIC COMMUNICATION, WHICH ARE VERY IMPORTANT TO SOLVE IMPLANTATION PROBLEMS IN ANIMALS OF HIGH GENETIC MERIT AND PRODUCTION, REPRODUCCION\EMBRION\OVIDUCTO\UTERO\VESICULAS EXTRACELULARES\EXOSOMAS\FERTILIDAD\VACUNO
