CLASSICALLY, MESENCHYMAL LINEAGE IS SHOWN AS "WADDINGTON DIAGRAM"-STYLE DIFFERENTIATION FLOW CHART, WITH A MESENCHYMAL STEM CELL (MSC) AT THE TOP OF THE HIERARCHY AND THEN PARALLEL LINEAGE COMMITMENTS, IN BONE CONTEXT, THE HIERARCHY AND ORIGIN OF BONE AND STROMA MESENCHYMAL CELLS YIELDED FROM ENDOCHONDRAL DIFFERENTIATION PROCESS HAS BEEN CONTROVERSIAL UNTIL RECENTLY, WITH OPPOSED HYPOTHESIS SUGGESTING THEY MAY ARISE FROM TRANSDIFFERENTIATION OF HYPERTROPHIC CARTILAGE OR FROM DIRECT DIFFERENTIATION OF BONE MARROW RESIDENT MSCS,IN THIS PROJECT, WE ARE WILLING TO ADDRESS THE MESENCHYMAL HIERARCHY IN BONE TISSUE CONTEXT AND ITS ROLE IN POSTNATAL PHYSIOLOGICAL PROCESSES, FOR THIS AIM, WE WILL TO USE TOMATO REPORTER IN CONDITIONAL CRE/LOX MICE TO PERFORM LINEAGE TRACING STUDIES OF GROWING PLATE CARTILAGE CELLS, TOMATO+ CELLS WILL BE HARVESTED FORM CARTILAGE AND BONE MARROW AREAS, PHENOTYPED AND DIFFERENTIATION POTENTIAL INTERROGATED WITHIN BIOMATERIALS-BASED 3D CELL CULTURE SYSTEMS, IN THIS SENSE, WE WILL TRY TO DEFINE THE CELL FATE UNDER DIFFERENT DIFFERENTIATION PROTOCOLS AND PICK UP TRANSDIFFERENTIATION STAGES AND REPROGRAMMING PROCESSES IN VITRO, ADDITIONALLY, WE WILL IMPLANT THESE 3D CELL CULTURES IN SECONDARY RECIPIENT MICE AND WE WILL PERFORM A SERIAL OF INTERVENTIONS TO DEFINE IN VIVO THE FATE OF CARTILAGE CELL PROGENY, FINALLY, WE WILL GENERATE CONDITIONAL CRE/LOX MICE MODELS BASED ON THE OUTCOME OF OUR IN VITRO MOLECULAR STUDIES AND AIMING TO DEFINE THE ROLE OF MESENCHYMAL CELL DYNAMICS IN BONE POSTNATALLY, MESENCHYMAL STEM CELLS\BONE\CARTILAGE\BIOMATERIALS
