TEMPERATURE IS ONE THE MOST IMPORTANT PARAMETERS IN MANY SCIENTIFIC FIELDS INCLUDING BIOLOGY, PHYSICS (SPECIFICALLY PHOTONICS), CHEMISTRY AND MEDICINE. IN BIOLOGY, THE INTRACELLULAR TEMPERATURE IS A KEY FACTOR IN CELL FUNCTION AS IT INFLUENCES SEVERAL CELL MECHANISMS AND PROCESSES. FOR THIS, THE POSSIBILITY OF ACCURATELY MEASURING AND MANIPULATE INTRACELLULAR TEMPERATURE ATTRACTS THE ATTENTION OF THE SCIENTIFIC COMMUNITY AS IS REFLECTED IN THE AMOUNT OF RESEARCH PUBLISHED ON THE TOPIC. ALTHOUGH BIG ADVANCES HAVE BEEN ACHIEVED IN ALL OF THESE FIELDS, THERE IS NO CURRENT RESEARCH FOCUSING ON UNDERSTANDING HOW TO BEST UTILIZE INTRACELLULAR TEMPERATURE MEASUREMENTS AND MANIPULATION FOR BIOLOGICAL APPLICATIONS. THIS PROPOSAL INTENDS TO DO JUST THAT ACCOMPLISHING TWO IMPORTANT OBJECTIVES:(1) CANCER DIAGNOSIS BASED ON INTRACELLULAR TEMPERATURE MEASUREMENTS AND IMAGING: TEMPERATURE IS CERTAINLY INVOLVED IN CANCEROGENESIS (CANCER CELLS WARMER THAN NON-CANCER CELLS). FOR THIS, A DEEPER UNDERSTANDING OF THE CONNECTION BETWEEN INTRACELLULAR TEMPERATURE AND INTRACELLULAR MECHANISMS AND PROCESSES MAY HELP TO UNDERSTAND THE ORIGIN OF CANCEROGENESIS. THIS UNDERSTANDING WILL PAVE THE WAY TO DEVELOP NEW BIOLOGICAL APPLICATIONS CENTERED IN INTRACELLULAR TEMPERATURE FOR CANCER RESEARCH. (2) IMPROVING ANTI-CANCER DRUG DELIVERY: THERAPEUTIC DRUG DELIVERY TO THE BRAIN REMAINS AN UNSOLVED CHALLENGE THAT LIMITS THE TREATMENT OF NEUROLOGICAL AND BRAIN DISEASES, SUCH AS GLIOBLASTOMA, PARKINSON, AND STROKE. THE MAIN LIMITATION IS IN THE ABILITY OF THERAPEUTIC DRUGS TO CROSS THE BLOOD-BRAIN BARRIER (BBB). TO OVERCOME THIS LIMITATION, SCIENTISTS PRESENT HYPERTHERMIA AS AN APPROACH TO OVERCOME THIS LIMITATION. ALTHOUGH THIS APPROACH SHOWS PROMISING RESULTS, THE STUDIES WERE NOT FOCUSED ON UNDERSTANDING AND COMBINING THE TEMPERATURE RAISE DURING THE BBB OPENING. THUS, THE POSSIBILITY OF USING THIS TEMPERATURE RISE TO BOTH IMPROVE THE BBB OPENING PERFORMANCE AND TO EXPLOIT IT TO TREAT BRAIN DISEASES IS NOT USED. FOR THE AFOREMENTIONED TO BE ACTUALIZED, THIS PROJECT WILL BRING THE LATEST ADVANCES IN INTRACELLULAR TEMPERATURE MEASUREMENTS AND IMAGING TO A DEEPER UNDERSTANDING OF THE ROLE OF TEMPERATURE IN THE BBB OPENING AND ITS POSSIBLE UTILIZATION IN THE TREATMENT OF BRAIN DISEASES, SUCH AS GLIOBLASTOMA. THUS, TWO IMPORTANT OBJECTIVES WILL BE ACHIEVED: (1) DETAILED STUDIES AND UNDERSTANDING OF BOTH THE INTRACELLULAR TEMPERATURE AND EXTRACELLULAR TEMPERATURE NEEDED FOR THE BBB OPENING AND (2) DESIGN AND SYNTHESIS OF THE NANO-COMPLEX FOR SIMULTANEOUSLY BBB OPENING AND THE RELEASE IN REAL-TIME AND ON-DEMAND OF THERAPEUTIC DRUGS FOR GLIOBLASTOMA TREATMENT IN VIVO. THESE GENERAL OBJECTIVES ARE MEANT TO PRESENT INTRACELLULAR TEMPERATURE AS A NEW PARADIGM IN MOLECULAR BIOLOGY UTILIZING THE LAST ADVANCES OF BIOPHOTONICS, MOLECULAR BIOLOGY, AND NANOTECHNOLOGY. IT IS EXPECTED THAT INTRACELLULAR TEMPERATURE WILL BECOME FULLY UTILIZED AND BE CONSIDERED A MEANINGFUL VARIABLE FOR MANY BIOLOGICAL APPLICATIONS. FROM THERE, IT IS EXPECTED THAT INTRACELLULAR TEMPERATURE WILL BECOME WIDELY USED IN MANY DIFFERENT RESEARCH PROJECTS AS IT WILL PROVIDE A MORE ACCURATE DESCRIPTION OF INTRACELLULAR ENVIRONMENTS THAN WHAT IS CURRENTLY AVAILABLE. NOT ONLY WILL THE OUTCOMES PRESENTED IN THIS PROPOSAL LEAD TO FURTHER INTRACELLULAR DISCOVERY AND UNDERSTANDING, BUT THEY WILL ALSO CREATE NEW HORIZONS IN SEVERAL DIFFERENT FIELDS OF RESEARCH.
