Descripción del proyecto
IT HAS BEEN SUGGESTED THAT DISRUPTIONS IN THE LONG-RANGE CONNECTIONS OF THE CEREBELLUM CONTRIBUTE TO THE PATHOGENESIS OF VARIOUS NEURODEVELOPMENTAL DISORDERS. INTERESTINGLY, THESE DISORDERS ARE CHARACTERIZED BY ATYPICAL CEREBELLAR DEVELOPMENT DURING CRITICAL PERIODS, INDICATING A DEVELOPMENTAL DIMENSION. THEREFORE, IT IS CRUCIAL TO UNDERSTAND HOW THESE LONG-RANGE CIRCUITS DEVELOP AND THE PRIMARY MECHANISMS DRIVING THEIR CORRECT WIRING IN ORDER TO COMPREHEND THE SPECIFIC ROLE OF THE CEREBELLUM IN BOTH TYPICAL AND ATYPICAL DEVELOPMENT. PARADOXICALLY OUR CURRENT KNOWLEDGE REGARDING THE ONTOGENY OF CEREBELLAR LONG-RANGE CONNECTIVITY AND THE UNDERLYING MECHANISMS REMAINS INSUFFICIENTLY EXPLORED. THE MAIN OBJECTIVE OF THIS PROPOSAL IS TO CHARACTERIZE THE DEVELOPMENTAL TRAJECTORIES OF THIS CIRCUITRY AND GAIN INSIGHTS INTO THE CONTRIBUTIONS OF EARLY CEREBELLAR SPONTANEOUS ACTIVITY TO THIS PROCESS. TO ACHIEVE THIS GOAL, WE WILL EMPLOY VARIOUS TECHNIQUES AND TOOLS TO SPECIFICALLY STUDY CEREBELLAR LONG-RANGE CONNECTIVITY AND MANIPULATE CEREBELLAR ACTIVITY DURING EARLY DEVELOPMENT, SUBSEQUENTLY ANALYZING THE ANATOMICAL AND TRANSCRIPTOMIC CONSEQUENCES OF SUCH ALTERATIONS. WE FIRMLY BELIEVE THAT OUR FINDINGS WILL SIGNIFICANTLY ENHANCE OUR UNDERSTANDING OF CEREBELLAR DEVELOPMENT, FUNCTION, AND DYSFUNCTION. CEREBELO\ACTIVIDAD\CIRCUITOS NEURONALES\DESARROLLO
