SUBMARINE GROUNDWATER DISCHARGE (SGD) I.E., THE DISCHARGE OF FRESH, BRACKISH OR SALINE GROUNDWATER FROM COASTAL AQUIFERS INTO THE OCEAN, IS A GLOBALLY IMPORTANT HYDROLOGIC PATHWAY THAT OCCURS ALONG MOST WORLD COASTLINES AND WHICH CAN HAVE PROFOUND IMPACTS IN COASTAL MARINE ECOSYSTEMS. COMPARED TO RIVERINE DISCHARGE, HOWEVER, SGD IS OFTEN INVISIBLE AND MUCH MORE SPATIALLY HETEROGENEOUS AND TEMPORALLY VARIABLE, EVEN AT VERY SMALL SCALES, WHICH POSE CHALLENGES FOR ITS QUANTIFICATION AND UNDERSTANDING ITS IMPACTS. CURRENTLY, ONE OF THE BIGGEST GAPS OF KNOWLEDGE IN THE FIELD OF SGD IS ITS BIOTIC DIMENSION, AS THE STUDY OF SGD HAS BEEN APPROACHED MOSTLY FROM A HYDROLOGICAL, OCEANOGRAPHICAL OR GEOCHEMICAL POINT OF VIEW, DISREGARDING ANY MICROBIAL ASPECTS LINKED TO THIS HYDROLOGIC PATHWAY. MICROBES PLAY KEY ROLES IN MODULATING SGD EFFECTS IN THE MARINE ENVIRONMENT BY CONTROLLING THE FLOW AND FATE OF THE GROUNDWATER CHEMICAL ELEMENTS BOTH ALONG COASTAL AQUIFERS AND ONCE IN THE OCEAN. HOWEVER, THESE MICROBIAL COMMUNITIES ARE ALSO EXTREMELY VARIABLE AT VERY SMALL SPATIAL SCALES, LEADING TO HIGHLY LOCALIZED HOTSPOTS OF MICROBIAL ACTIVITY IN COASTAL GROUNDWATER AS WELL AS TO A PATCHY AND DIFFICULT TO PREDICT DISTRIBUTION OF SGD QUANTITY, QUALITY, AND THE ASSOCIATED RESPONSES OF MARINE MICROBES. IN THIS CONTEXT, THE PATCHI PROJECT WILL AIM AT CHARACTERIZING THE MICROBIAL ASPECTS OF SGD AT SMALL SPATIAL SCALES THAT HAVE NOT BEEN CONSIDERED PREVIOUSLY (ON THE ORDER OF CENTIMETERS/METERS), BOTH WITHIN THE COASTAL AQUIFER NEXT TO THE SHORELINE AND IN THE SEA RIGHT AT THE DISCHARGE POINTS. THROUGH A COMBINATION OF DETAILED CROSS-SHORE TRANSECT POREWATER SAMPLINGS IN DIFFERENT MEDITERRANEAN BEACHES AND IN SITU NATURAL GROUNDWATER ADDITION EXPERIMENTS USING SEEPAGE METERS (I.E., SGD COLLECTION CHAMBERS), PATCHI WILL AIM AT DESCRIBING IN DETAIL THE SPATIAL VARIABILITY IN TAXONOMIC COMPOSITION AND METABOLIC FUNCTION OF I) THE GROUNDWATER MICROBES DRIVING CHANGES IN SGD AND II) THE MARINE MICROBES EXPOSED TO SPATIALLY VARIABLE SGD. METAGENOMIC AND AMPLICON SEQUENCING WILL BE COUPLED WITH FLOW CYTOMETRY AND ACTIVITY ASSAYS FOR A COMPLETE DESCRIPTION OF THESE POORLY EXPLORED MICROBIAL COMMUNITIES. OUR RESULTS WILL BE COMPARED WITH BROADER-SCALE INTEGRATED SGD ESTIMATES SUCH AS THOSE BASED ON RADON OR RADIUM ISOTOPIC TRACERS. ALTOGETHER, PATCHI WILL ESTABLISH OR REFINE THE SCALE AT WHICH WE SHOULD STUDY SGD, AT LEAST IN THE MEDITERRANEAN SEA, DELINEATING THE EXTENT TO WHICH GROUNDWATER DISCHARGE AFFECTS THE STRUCTURE AND FUNCTIONING OF THE MARINE BENTHIC AND PLANKTONIC MICROBIOME.
