THE ASCOMYCETOUS FAMILY MAGNAPORTHACEAE INCLUDES PLANT PATHOGENIC FUNGI THAT CAUSE DISEASES ON MORE THAN 50 MEMBERS OF THE POACEAE WORLDWIDE, AFFECTING THE TURFGRASS INDUSTRY AND STAPLE FOOD CROPS SUCH AS RICE, WHEAT, MILLET AND MAIZE. THE ECONOMICALLY SIGNIFICANT TURFGRASS ROOT PATHOGEN MAGNAPORTHE POAE BELONGS TO THIS FAMILY. TWO OF THE MOST DAMAGING FUNGAL PATHOGENS OF WHEAT AND RICE ALSO BELONG TO THIS FAMILY: THE INTRACTABLE ROOT-INFECTING FUNGUS AND CAUSAL AGENT OF TAKE-ALL DISEASE, GAEUMANNOMYCES GRAMINIS AND THE BLAST FUNGUS MAGNAPORTHE ORYZAE. M. ORYZAE HAS EMERGED AS A PARADIGM FOR THE UNDERSTANDING OF FUNGAL AERIAL ATTACK. INTERESTINGLY, M. ORYZAE CAN ALSO INFECT ROOTS (UNDER LABORATORY CONDITIONS) AND IS FAR MORE AMENABLE TO TRANSFORMATION THAN G. GRAMINIS AND M. POAE. VERY LITTLE IS KNOWN ABOUT THE REQUIREMENTS OF SOIL-BORNE FUNGAL PATHOGENS FOR SUCCESSFUL COLONISATION OF ROOTS. CONSEQUENTLY, M. ORYZAE-RICE INTERACTION CONSTITUTES AN EXCELLENT PATHOSYSTEM FOR THE IDENTIFICATION OF DEVELOPMENTAL AND METABOLIC REQUIREMENTS FOR INFECTION OF UNDERGROUND TISSUES, AND FOR COMPARATIVE STUDIES OF THE DISTINCT ORGAN-SPECIFIC PATHOGENIC MECHANISMS OBSERVED DURING LEAF AND ROOT COLONISATION. M. ORYZAE MITOGEN-ACTIVATED PROTEIN KINASE (MAPK) PMK1 GENE AND ITS ORTHOLOGUES HAVE BEEN FOUND TO REGULATE DEVELOPMENT AND VIRULENCE IN ALL FUNGAL PLANT PATHOGENS STUDIED TO DATE. TRANSCRIPTION FACTORS CONSTITUTE ONE OF THE DOWNSTREAM TARGETS OF MAPK SIGNALLING CASCADES. THE TRANSLOCATION OF TRANSCRIPTION FACTORS BETWEEN NUCLEUS AND CYTOPLASM IS CONTROLLED BY PROTEIN RECEPTORS CALLED KARYOPHERINS. WE HAVE CHARACTERISED A M. ORYZAE MUTANT WHICH LACKED THE M. ORYZAE ORTHOLOG OF THE KARYOPHERIN EXPORTIN-5/MSN5P (EXP5). THE EXP5 MUTANTS WERE MUCH LESS VIRULENT ON ROOTS, SUGGESTING AN IMPORTANT INVOLVEMENT OF PROTEINS AND/OR RNAS TRANSPORTED BY EXP5 DURING M. ORYZAE ROOT INFECTION. EXP5 ALSO TRANSPORT DOUBLE STRANDED RNA-BINDING PROTEINS AND NON-CODING RNAS INCLUDING PRECURSORS OF MIRNAS TO THE CYTOPLASM. AT PRESENT THERE IS A LACK OF KNOWLEDGE OF THE EXP5-DEPENDENT CARGOES IN FILAMENTOUS FUNGI.OBJECTIVES OF PROPOSED RESEARCH PROGRAMME THE AIM OF THIS PROJECT IS TO INVESTIGATE FUNDAMENTAL MOLECULAR MECHANISMS THAT REGULATE THE ABILITY OF M. ORYZAE TO INFECT RICE. THE WORK OUTLINED IN THIS PROPOSAL WILL EXTEND THIS AREA OF RESEARCH THROUGH I) A BETTER UNDERSTANDING OF THE CELLULAR AND METABOLIC MECHANISMS UNDERLYING ORGAN-SPECIFICITY IN M. ORYZAE USING A COMPARATIVE TRANSCRIPTOME ANALYSIS, AND II) THE IDENTIFICATION OF EXP5-DEPENDENT CARGOES AND FUNCTIONAL ANALYSIS OF THOSE INVOLVED IN M. ORYZAE RICE COLONISATION.TARGETS(1) RNA EXTRACTIONS FROM INFECTED LEAVES AND ROOTS AND MICROARRAY HYBRIDIDATIONS BY YEAR 1. ANALYSIS OF DATA AND IDENTIFICATION OF FUNGAL METABOLIC PATHWAYS AND CELLULAR PROCESSES BY YEAR 2. MUTANT GENERATION BY TARGETED GENE REPLACEMENT OF SELECTED GENE CANDIDATES (1 OR 2) AND THEIR FUNCTIONAL CHARACTERISATION BY YEAR 3. (2) GENERATION OF A FUNCTIONAL HA-FLAG-TAGGED EXP5 BY YEAR1. IDENTIFICATION OF EXP5-DEPENDENT PROTEINS AND NORTHERN BLOTS USING SUBCELLULAR FRACTIONS BY YEAR 2. IDENTIFICATION OF EXP5-DEPENDENT RNA CARGOES BY YEAR 3. TARGETED GENE REPLACEMENT OF ONE SELECTED EXP5-DEPENDENT CARGO AND INITIATION OF ITS MECHANISTIC STUDIES BY YEAR 3.
